Regarding disabling spanning tree; if you have spanning tree disabled, then it is possible that layer 2 loops will form in your network. This will cause issues with (for example) broadcast traffic, as Ethernet does not have any concept of 'Time to live'; broadcast frames will loop around the network forever, slowly eating up resources.

Use Multi-Spanning Tree Protocol (MST/MSTP) instead of RSTP if you have a green field network and your devices support it. It scales better and requires less CPU as the number of VLANs increase. Configure your aggregation switches (looks like a collapsed core) (RSW) with the lowest STP bridge priorities. Choose one as the lowest and root.

The optimisations and changes that you're proposing are moot if the topology of the network won't be changing anyway. Spanning-tree is a low impact protocol in the first place. 600 ports / stations isn't that much in the big scheme of things considering spanning-tree.
