Ribonuclease (commonly abbreviated RNase) is a type of nuclease that catalyzes the degradation of RNA into smaller components. Ribonucleases can be divided into endoribonucleases and exoribonucleases, and comprise several sub-classes within the EC 2.7 (for the phosphorolytic enzymes) and 3.1 (for the hydrolytic enzymes) classes of enzymes.

Bovine pancreatic ribonuclease A (RNase A) catalyzes the cleavage of P–O 5′ bonds in RNA. Structural analyses had suggested that the active-site lysine residue (K41) may interact preferentially with the transition state for covalent bond cleavage, thus facilitating catalysis.

2005年12月5日 · To determine the sequence of ribonuclease, Moore, Stein, and their first postdoctoral associate, Werner Hirs, hydrolyzed the protein with trypsin and then separated the peptide mixture by ion-exchange chromatography. The peptide sequences were then analyzed by Edman degradation.

RNase A is a 14 kDa monomeric enzyme that catalyzes the transphosphorylation of single stranded RNA with specificity for pyrimidines on the 3′ side of the bond to be cleaved and purines on the 5′ side of this bond.
