You can take public transport to within about 10k of the sanctuary, but after that you need to arrange a car. We found it extremely easy to reserve an Uber to pick us up at the …
You can take public transport to within about 10k of the sanctuary, but after that you need to arrange a car. We found it extremely easy to reserve an Uber to pick us up at the sanctuary at the end of the tour, and to get an Uber from Frankston or Cranbourne stations (either one is the closest - they are on separate train limes).

I don’t think it’s logistically possible for one day. If I recall correctly, both start at sunset. And the penguin one you need to be there a good deal beforehand to get a good spot, etc. …
